Technique basics
ISH is the process by which the specific annealing of a labelled nucleic acid to complementary sequences in fixed tissues or cells is followed by visualization of the hybridized probe
This hybridization reaction between a labelled nucleotide probe and a complementary DNA or RNA sequence is visualized directly on its target, which provides assertive interpretations while preserving integrity of the cells.
The annealing and separation of the two hybridized strands depend on various factors that are controlled according to the in situ Hybridization protocol in order to elevate the chances of a good hybridization occurring.
